Abstract
The present invention relates to antisense oligonucleotides that modulate the expression of and/or function of Tumor Suppressor genes, in particular, by targeting natural antisense polynucleotides of Tumor Suppressor genes. The invention also relates to the identification of these antisense oligonucleotides and their use in treating diseases and disorders associated with the expression of Tumor Suppressor genes.
Claims14
A single-stranded synthetic, modified oligonucleotide of 15 to 27 nucleotides in length comprising at least one modification, wherein the at least one modification is selected from:
- at least one modified sugar moiety;
- at least one modified internucleotide linkage;
- at least one modified nucleotide;
- and combinations thereof;
- and further wherein said oligonucleotide is an antisense compound which is 100% complementary to and specifically hybridizes to a target region of a human natural antisense polynucleotide selected from the group consisting of SEQ ID NOs:
- 14 or 15, and wherein the oligonucleotide upregulates expression and/or function of a human PTEN gene polynucleotide having SEQ ID NO:
- 6 in vivo or in vitro as compared to a normal control oligonucleotide, wherein the target region on SEQ ID NO:
- 14 is selected from nucleotides 1-75, 137-159, 161-191, 196-301, 317-337 and 399-789 on said SEQ ID NO:
- 14.
