Abstract
Disclosed are compositions and methods for directing NK cells to the bone marrow through the use of PM21 particles.
Claims7
A method of trafficking natural killer (NK) cells to bone marrow comprising contacting NK cells ex vivo with one or more of plasma membrane particles expressing IL-21 and 41 BBL on their surface (PM21 particles) and/or feeder cells expressing IL-21 on their surface (FC21 feeder cells) for 6 to 40 days, wherein the contacting induces a cellular mechanism within NK cells to induce fucosylation of P-selectin glycoprotein ligand-1 (PSGL-1) on the NK cells' surface.
A method of treating a bone marrow malignancy or bone marrow born malignancy in a subject comprising contacting NK cells ex vivo with plasma membrane particles expressing IL-21 and 41 BBL on their surface (PM21 particles) and/or feeder cells expressing IL-21 on their surface (FC21 feeder cells) for 6 to 40 days, wherein the contacting induces a cellular mechanism within NK cells to induce fucosylation of P-selectin glycoprotein ligand-1 (PSGL-1) on the NK cells' surface, and adoptively transferring the NK cells to the subject.
