Abstract
The present disclosure provides automated modules and instruments for improved detection of nuclease genome editing of live cells. The disclosure provides improved modules—including high throughput modules—for screening cells that have been subjected to editing and identifying and selecting cells that have been properly edited.
Claims10
A cell editing system comprising:
- a robotic handling instrument;
- a growth component having a growth chamber to grow cells;
- a transformation component having a transformation chamber for receiving the cells from the growth component and for receiving nucleic acids, to create transformed cells;
- an editing component having a first chamber to receive the transformed cells, to substantially singulate the transformed cells into cell colonies;
- a storage component;
- and
- a processor operably coupled to the robotic handling instrument, the processor being configured to execute a script to:
- transfer the cells from the growth component to the transformation component in response to a first signal from a first sensor;
- transfer the nucleic acids to the transformation component in response to a second signal from a second sensor;
- transfer the transformed cells to the editing component in response to a third signal from a third sensor;
- and
- transfer a first cell colony, of the cell colonies, to the storage component in response to a fourth signal from a fourth sensor.
