Abstract
Described herein are polypeptides, systems, and methods that relate to using domains that bind specifically to a biotinylamide to control receptor and cellular activity.
Claims23
A synthetic signaling system comprising:
- a small molecule-controlled signaling polypeptide comprising
- i) a small molecule acceptor peptide and
- ii) at least a first signaling domain,
- wherein the i) a small molecule acceptor peptide and ii) at least a first signaling domain of the small molecule-controlled signaling polypeptide are not found as part of the same polypeptide in nature;
- and
- a polypeptide comprising a domain that binds specifically to the small molecule;
- and
- a surface-attached molecule comprising:
- a) a binding domain specific for a target, and
- b) a small molecule acceptor peptide.

A combination comprising:
- I) a cell or set of cells comprising one or more nucleic acids encoding:
- A) one of:
- i) a small molecule-controlled signaling polypeptide comprising:
- 1) A small molecule acceptor peptide and
- 2) at least a first signaling domain;
- wherein the 1) a small molecule acceptor peptide and 2) at least a first signaling domain of the small molecule-controlled signaling polypeptide are not found as part of the same polypeptide in nature;
- and
- ii) a small molecule-controlled signaling polypeptide comprising at least a first signaling domain;
- and
- B) a polypeptide comprising a domain that binds specifically to the small molecule;
- and
- II) a surface-attached molecule comprising:
- a) a binding domain specific for a target, and
- b) a small molecule acceptor peptide.

A combination comprising:
- I) a cell or set of cells comprising a nucleic acid encoding a small molecule-controlled signaling polypeptide, the small molecule-controlled signaling polypeptide comprising i) a small molecule acceptor peptide and ii) at least a first signaling domain;
- and
- II) a surface-attached molecule comprising:
- a) a binding domain specific for a target, and
- b) a small molecule acceptor peptide.

A method of treating a subject in need of immunotherapy, the method comprising administering to the subject an immune cell comprising a synthetic signaling system comprising:
- a small molecule-controlled signaling polypeptide comprising
- i) a small molecule acceptor peptide and
- ii) at least a first signaling domain,
- wherein the i) a small molecule acceptor peptide and ii) at least a first signaling domain of the small molecule-controlled signaling polypeptide are not found as part of the same polypeptide in nature;
- and
- a polypeptide comprising a domain that binds specifically to the small molecule.
