Abstract
The present disclosure provides compositions and method targeting GPER for the treatment of cancers, such as breast cancers and leukemias, gallstone disease, and for conferring of neuroprotection on a subject. Also disclosed are high throughput assays for identifying antagonists of GPER.

A compound of the formula:
- wherein:
- R1 is cycloalkyl(C≤12) substituted cycloalkyl(C≤12);
- R2 is alkyl(C≤12), cycloalkyl(C≤12), aryl(C≤12), or a substituted version of any of these groups;
- R3 and R4 are each independently hydrogen, amino, halo, or hydroxy, or alkyl(C≤12), cycloalkyl(C≤12), aryl(C≤12), alkoxy(C≤12), or a substituted version of any of these groups;
- or
- R3 and R4 are taken together and are alkenediyl(C≤8) and form a second aromatic ring;
- R5 is hydrogen, alkyl(C≤6), or substituted alkyl(C≤6);
- m is 0, 1, 2, or 3;
- and
- n is 1 or 2;
- or a pharmaceutically acceptable salt thereof.
