Abstract
Oral formulations comprising a compound of Formula (I) and a chelating agent are provided, wherein compounds of Formula (I) have the structure: wherein, W, X, Y, Z, Ra, Rc, and each Rg are as defined herein. Also provided are methods of preparation, methods of use, and specific dosage forms.
Claims24
An oral formulation comprising a compound of Formula (I) or a pharmaceutically acceptable salt, hydrate or solvate thereof, and inositol hexaphosphate or an ionized salt thereof, wherein the compound of Formula (I) has the structure:
- wherein,
- W is selected from the group consisting of CRe and N;
- X is selected from the group consisting of O, CH2, and S;
- each of Y and Z is independently selected from the group consisting of CH and N:
- Rg is H or the two R9 groups are combined to form an acetonide;
- Ra is selected from the group consisting of NH2, NHR1, and NR1R2;
- Rc is selected from the group consisting of H, halogen, haloalkyl, NH2, NHR3, NR3R4, R3, OH, OR3, SR3, SO2R3, —X1—NH2, —X1—NHR3, —X1—NR3R4, —X1—OH, —X1—OR3, —X1—SR3, and —X1—SO2R3;
- Re is selected from the group consisting of H, halogen, and optionally substituted C1-C6 alkyl;
- each X1 is C1-C4alkylene;
- and
- each R1, R2, R3, and R4 is independently selected from the group consisting of optionally substituted C1-C10 alkyl, optionally substituted C3-C7 cycloalkyl, optionally substituted C3-C7 cycloalkylC1-C4alkyl-, optionally substituted 4-7 membered heterocycloalkyl, optionally substituted 4-7 membered heterocycloalkylC1-C4alkyl-, optionally substituted aryl, optionally substituted arylC1-C4alkyl-, optionally substituted heteroaryl, and
- optionally substituted heteroarylC1-C4alkyl-, or when R1 and R2 or R3 and R4 are attached to the same nitrogen, they combine to form a 4- to 7-membered heterocyclic ring.
An oral formulation comprising a compound of Formula (I) or a pharmaceutically acceptable salt, hydrate or solvate thereof, and edetic acid (EDTA) or an ionized salt thereof, wherein the compound of Formula (I) has the structure:
- wherein,
- W is selected from the group consisting of CRe and N;
- X is selected from the group consisting of O, CH2, and S;
- each of Y and Z is independently selected from the group consisting of CH and N:
- Rg is H or the two Rg groups are combined to form an acetonide;
- Ra is selected from the group consisting of NH2, NHR1, and NR1R2;
- Rc is selected from the group consisting of H, halogen, haloalkyl, NH2, NHR3, NR3R4, R3, OH, OR3, SR3, SO2R3, —X1—NH2, —X1—NHR3, —X1—NR3R4, —X1—OH, —X1—OR3, —X1—SR3, and —X1—SO2R3;
- Re is selected from the group consisting of H, halogen, and optionally substituted C1-C6 alkyl;
- each X1 is C1-C4alkylene;
- and
- each R1, R2, R3, and R4 is independently selected from the group consisting of optionally substituted C1-C10 alkyl, optionally substituted C3-C7 cycloalkyl, optionally substituted C3-C7 cycloalkylC1-C4alkyl-, optionally substituted 4-7 membered heterocycloalkyl, optionally substituted 4-7 membered heterocycloalkylC1-C4alkyl-, optionally substituted aryl, optionally substituted arylC1-C4alkyl-, optionally substituted heteroaryl, and optionally substituted heteroarylC1-C4alkyl-, or when R1 and R2 or R3 and R4 are attached to the same nitrogen, they combine to form a 4- to 7-membered heterocyclic ring.
