Abstract
The present disclosure provides oligomeric compounds (including oligomeric compounds that are antisense agents or portions thereof) comprising a modified oligonucleotide having at least one modified internucleoside linking group.
Claims28
An oligomeric compound comprising a modified oligonucleotide consisting of 12-22 linked nucleosides, wherein the modified oligonucleotide has a 5′-region, a central region, and a 3′-region, wherein:
- the 5′-region consists of 1-6 linked nucleosides, each comprising a modified sugar moiety;
- the 3′-region consists of 1-6 linked nucleosides, each comprising a modified sugar moiety;
- the central region is a deoxy region;
- and
- the central region consists of 7-12 linked nucleosides and has the formula (from 5′ to 3′):
- (Nd1)L1(Nd2)L2(Nd3)L3(Nd4)L4[(Nd)L5]q, wherein:
- Nd1, Nd2, Nd3, Nd4 are independently selected from a stereo-standard DNA nucleoside and a 2′-substituted nucleoside;
- with the proviso that no more than one of Nd1, Nd2, Nd3, or Nd4 is a 2′-substituted nucleoside;
- q is from 3-8;
- each of L1, L2, L3, L4, and each L5 is an internucleoside linkage:
- two, three, or four of L1, L2, L3, and L4 are mesyl phosphoramidate internucleoside linkages;
- and
- each L5 is a phosphorothioate internucleoside linkage.
