Abstract
Provided herein are methods for enhancing the transduction efficiency of vectors into cells, e.g., primary human T lymphocytes.
Claims22
A method of transducing a primary T lymphocyte, comprising:
- (i) contacting a primary T lymphocyte with a compound that is an inhibitor of the innate immune system and inhibits the activity of protein kinase R (PKR);
- and
- (ii) contacting the primary T lymphocyte with a viral vector comprising a nucleic acid;
- wherein the contacting of the primary T lymphocyte with the compound occurs prior to the contacting of the primary T lymphocyte with the viral vector;
- whereby the nucleic acid is transduced into the primary T lymphocyte.

A method of transducing a human primary T lymphocyte, comprising:
- (i) contacting a human primary T lymphocyte, in the presence of diethylaminoethyl-dextran and interleukin 2 (IL-2), with 2-aminopurine;
- and
- (ii) contacting the human primary T lymphocyte with a lentiviral vector comprising a nucleic acid encoding a chimeric antigen receptor;
- wherein the contacting of the human primary T lymphocyte with the 2-aminopurine occurs prior to the contacting of the human primary T lymphocyte with the lentiviral vector;
- whereby the nucleic acid is transduced into the T lymphocyte.
