Abstract
Recombinant DNA constructs, for use in plants and plant cells, have site-specific recombination sites that allow assessing phenotypes and modes of action by over expression or suppression of endogenous genes. In an aspect, a single DNA construct can be switched between over expression and suppression by the action of a recombinase such as the Cre recombinase on constructs having lox recombination sites. Other useful recombination systems include the Flp/frt system, the R/Rs system, the Dre/rox system, and the GIN/gix system.
Claims15
A recombinant DNA construct comprising:
- (A) a promoter operably linked to a first DNA sequence that is complementary to at least a fragment of a second DNA sequence, wherein the first DNA sequence is at least 21 nucleotides in length, and wherein the first DNA sequence is in anti-sense orientation with respect to the promoter and second DNA sequence;
- (B) the second DNA sequence, wherein the second DNA sequence encodes a protein and is in sense orientation with respect to the promoter;
- (C) at least one DNA sequence providing a polyadenylation sequence;
- (D) a first recombination site and a second recombination site, wherein the first and second recombination sites are recognized by a site-specific recombinase, wherein the first recombination site is positioned between the promoter and the first DNA sequence, and wherein the second recombination site is positioned between the first DNA sequence and the second DNA sequence.
