An isolated CRISPR from Prevotella and Francisella 1 (Cpf1) protein from Acidaminococcus sp. BV3L6 (AsCpf1), comprising an amino acid sequence having at least 95% sequence identity to the amino acid sequence of SEQ ID NO:
- 2, and further comprising an amino acid mutation, and the mutation is E174R.
