Abstract
Provided herein are automated apparatus for the identification of microorganisms in various samples. The disclosure solves existing challenges encountered in identifying and distinguishing various types of microorganisms, including viruses and bacteria in a timely, efficient, and automated manner by sequencing.
Claims21
A nucleic acid sequencing apparatus comprising:
- (a) a compartment configured to contain a plurality of nucleic acids for a nanopore sequencing reaction, wherein the compartment comprises two or more containers that are mechanically coupled, wherein each container of the two or more containers comprises a subset of nucleic acids from the plurality of nucleic acids;
- (b) a nucleic acid sequencing chamber operatively coupled to the compartment, wherein the nucleic acid sequencing chamber comprises a flow cell within the nucleic acid sequencing chamber, wherein the flow cell comprises:
- (i) a sample input port comprising a flow path in fluid communication with an outlet of the nucleic acid sequencing chamber, wherein the sample input port is configured to be robotically opened;
- and
- (ii) a plurality of nanopores configured for passage of the subset of nucleic acids from the sample input port to the outlet;
- (c) a robotic arm comprising a pipette, wherein the pipette comprises a fluid pump configured for (i) pumping a reagent into the two or more containers, and (ii) pumping the subset of nucleic acids from the plurality of nucleic acids into the flow cell;
- and
- (d) a computing device in communication with the robotic arm, wherein the computing device comprises:
- a memory location comprising instructions configured to perform transferring, by the robotic arm, the subset of nucleic acids from the two or more containers of the compartment into the sample input port of the flow cell within the nucleic acid sequencing chamber;
- wherein the transferring is performed under conditions sufficient to prevent failure of the flow cell due to introduction of air into the flow path of the flow cell.
