Abstract
The present invention relates to excision of explant material comprising meristematic tissue from seeds, and storage of such material prior to subsequent use in plant tissue culture and genetic transformation. Methods for tissue preparation, storage, and transformation are disclosed, as is transformable meristem tissue produced by such methods, and apparati for tissue preparation.
Claims27
A method for transforming a plant comprising:
- (a) preparing an explant from a plant seed under conditions wherein the explant does not germinate and remains viable and competent for genetic transformation;
- wherein the explant comprises meristematic tissue, and wherein the explant comprises an internal moisture content of from about 3% to about 25%;
- and
- (b) transforming at least a first cell of the explant with a selected DNA.
A method for transforming a plant comprising:
- (a) obtaining an explant from a plant seed, wherein the explant comprises meristematic tissue;
- (b) dehydrating the explant, wherein the explant after dehydrating does not germinate and remains viable and competent for genetic transformation and comprises an internal moisture content of from about 3% to about 25%;
- and
- (c) transforming at least a first cell of the explant with a selected DNA.
A method for producing a transformable plant tissue comprising:
- (a) obtaining a regenerable explant from a plant seed, wherein the explant comprises meristematic tissue;
- and
- (b) dehydrating the explant to obtain a dried and regenerable explant comprising an internal moisture content of from about 3% to about 25%.
