Abstract
De novo synthesized large libraries of nucleic acids are provided herein with low error rates. Further, devices for the manufacturing of high-quality building blocks, such as oligonucleotides, are described herein. Longer nucleic acids can be synthesized in parallel using microfluidic assemblies. Further, methods herein allow for the fast construction of large libraries of long, high-quality genes. Devices for the manufacturing of large libraries of long and high-quality nucleic acids are further described herein.
Claims16
A method of polynucleotide synthesis, comprising:
- (a) providing preselect sequences for a plurality of polynucleotides;
- (b) providing an assembly comprising:
- i. a reactor comprising a substrate, wherein the substrate is configured to hold liquid in a plurality of microwells;
- and
- ii. a vacuum chuck, wherein the vacuum chuck is in between the substrate and a temperature control device;
- wherein the substrate is aligned with an inkjet assembly configured to deposit one or more droplets;
- and
- (c) synthesizing the plurality of polynucleotides at the plurality of microwells by controlling a reaction condition, wherein the reaction condition comprises a temperature of at least 25 degrees Celsius;
- and
- (d) cleaving the plurality of polynucleotides from the plurality of microwells.
