Abstract
The present invention relates to excision of explant material comprising meristematic tissue from seeds, and storage of such material prior to subsequent use in plant tissue culture and genetic transformation. Methods for tissue preparation, storage, and transformation are disclosed, as is transformable meristem tissue produced by such methods, and apparati for tissue preparation.
Claims30
A method for obtaining transformed plant tissue comprising:
- (a) transforming at least a first cell of an explant with a selected DNA;
- and
- (b) regenerating a transgenic plant from said cell, wherein the plant is stably transformed with the selected DNA;
- further wherein the explant is prepared from a plant seed and comprises a meristem of the plant seed;
- and wherein the explant comprises an internal moisture content of from about 4% to about 16% prior to transforming the explant in step (a).
