Abstract
The present invention generally relates to droplet libraries and to systems and methods for the formation of libraries of droplets. The present invention also relates to methods utilizing these droplet libraries in various biological, chemical, or diagnostic assays.
Claims16
An emulsion library comprising:
- a plurality of stable aqueous droplets within an immiscible oil comprising at least one surfactant, wherein each droplet is uniform in size and most of the droplets comprise:
- one bead comprising an optical label specific for a target, wherein the bead is linked to a first antibody that binds to the target;
- and
- a second enzyme-linked antibody that binds to the target,
- wherein the droplets do not include the target and do not coalesce during storage and further wherein the oil and the surfactant have been selected to prevent the exchanges of contents among the droplets during the storage, wherein the emulsion library includes beads labeled with a plurality of optical labels, wherein each of the plurality of optical labels comprises a dye of increasing concentration from one label to the next.

- wherein the droplets do not include the target and do not coalesce during storage and further wherein the oil and the surfactant have been selected to prevent the exchanges of contents among the droplets during the storage, wherein the aqueous droplets have been stored have been stored for a period greater than 90 days under conditions that tend to promote droplet coalescence and wherein the droplets have a stable shelf life and do not coalesce for the period greater than 90 days.

An emulsion library comprising:
- a plurality of stable aqueous droplets within an immiscible oil comprising at least one surfactant, wherein each droplet is uniform in size and most droplets comprise:
- exactly one bead comprising an optical label specific for a target, wherein the bead is linked to a first antibody that binds to the target;
- and
- a second antibody that binds to the target, wherein the second antibody is linked to streptavidin or biotin, wherein the emulsion library is useful for an ELISA assay to detect an antigen from a sample but the droplets do not contain the antigen or the sample and further wherein the emulsion library includes beads labeled with a plurality of optical labels, wherein each of the plurality of optical labels comprises a dye of increasing concentration from one label to the next.
