Abstract
The present invention provides for heterologous expression of polypeptides encoded by wild-type and condon-optimized variants of cbh1 and/or cbh2 from the fungal organisms Talaromyces emersonii (T. emersonii), Humicola grisea (H. grisea), Thermoascus aurantiacus (T. aurantiacus), and Trichoderma reesei (T. reesei) in host cells, such as the yeast Saccharomyces cerevisiae. The expression in such host cells of the corresponding genes, and variants and combinations thereof, result in improved specific activity of the expressed cellobiohydrolases. Thus, such genes and expression systems are useful for efficient and cost-effective consolidated bioprocessing systems.
Claims13
A yeast host cell comprising a heterologous polynucleotide encoding a cellobiohydrolase polypeptide having 100% sequence identity with the polypeptide of SEQ ID NO:
- 11, wherein the heterologous polynucleotide comprises a codon-optimized nucleic acid sequence, wherein the codon-optimized nucleic acid sequence comprises the nucleic acid sequence of SEQ ID NO:
- 7 wherein the codon adaptation index (CAI) of the codon-optimized nucleic acid molecule is about 0.8 to about 1.0, wherein said CAI is determined using the codon values specific for the yeast host cell species;
- and wherein the cellobiohydrolase, expressed by the yeast host cell from the codon-optimized nucleic acid sequence, is capable of hydrolyzing Avicel.
A yeast host cell comprising a vector, wherein said vector comprises a heterologous polynucleotide encoding a cellobiohydrolase polypeptide having 100% sequence identity with the polypeptide of SEQ ID NO:
- 11, wherein the heterologous polynucleotide comprises a codon-optimized nucleic acid sequence, wherein the codon-optimized nucleic acid sequence comprises the nucleic acid sequence of SEQ ID NO:
- 7,wherein the codon adaptation index (CAI) of the codon-optimized nucleic acid molecule is about 0.8 to about 1.0, wherein said CAI is determined using the codon values specific for the yeast host cell species, and wherein the cellobiohydrolase, expressed by the yeast host cell from the codon-optimized nucleic acid sequence, is capable of hydrolyzing Avicel.
