Abstract
The present invention relates to a series of substituted purine derivatives capable of inhibiting CDC7 kinase activity and, as such, suitable for use in the treatment of neurological diseases such as, inter alia, Alzheimer's disease, amyotrophic lateral sclerosis or frontotemporal dementia, involving hyperphosphorylation of TDP-43 and the subsequent formation of aggregates, induced by CDC7.
Claims9
A method for the inhibition of CDC7 in the phosphorylation of TDP-43 for the treatment of pathologies related to the protein TDP-43, wherein the TDP-43 related disease is a neurological disease selected from amyotrophic lateral sclerosis, frontotemporal dementia, Alzheimer's disease, age-associated cognitive impairment, and chronic traumatic encephalopathy, the method comprising administering to a subject an effective amount of a compound according to formula (II):
- wherein R5 is H and at least one of R1, R2, R3 or R4 is Cl, Br, I, methyl, CF3, CN or NO2;
- and
- Z is selected from O or S.
A compound selected from:
- 6-((3-chlorobenzyl)oxy)-9H-purine,
- 6-((3-iodobenzyl)thio)-9H-purine, and
- 6-((4-bromobenzyl)oxy)-9H-purine.
