Abstract
An organ-on-a-chip microfluidic device is disclosed that mimics a human lymph node and/or human lymphoid tissue. The device can include cells from human blood and lymphatic tissue, include an extracellular matrix for the development of immune system components, and provide for the perfusion of fluids and solids resembling blood and lymphatic fluid within micrometer sized channels.
Claims19
A method comprising:
- a) providing a microfluidic device comprising a body, said body comprising a fluid inlet and a fluid outlet, said fluid inlet and fluid outlet in fluidic communication with a first microchannel, said first microchannel in fluidic communication with a first chamber, the first chamber comprising a matrix;
- b) providing T lymphocytes and B lymphocytes within the matrix, wherein the density of said T lymphocytes and said B lymphocytes within the matrix is greater than 500,000 cells per milliliter;
- and
- c) culturing said T lymphocytes and said B lymphocytes wherein the culturing comprises perfusing said T lymphocytes and said B lymphocytes with culture media at a flow rate to produce B lymphocyte clusters within the matrix and wherein the B lymphocyte clusters contain less than 10% T lymphocytes.
