NSD family inhibitors and methods of treatment therewith

A composition comprising a compound of Formula (IIb):

• wherein X is CH2 or NH;
• wherein R2 is NH2, C1-C4 alkyl, C1-C4 haloalkyl, C1-C4 dihaloalkyl, C1-C4 trihaloalkyl, and C1-C4 aminoalkyl;
• wherein R4 is selected from H, halogen, OH, OCH3, OCH2CH3, CH2OH, NH2, CH2NH3, NHCH3, CH3, CH2CH3, CHCH2, trihalomethyl, trihaloethyl, dihalomethyl, dihaloethyl, halomethyl, and haloethyl;
• wherein R5 is selected from H, CH3, CH2CH3, OH, OCH3, NH2, and halogen;
• wherein Z is NH, NCH3, S, O, CH2, or is absent;
• wherein A is selected from a six-member aryl or heteroaryl ring, a heteroaryl bicyclic ring system comprising a five-member ring and a six-member ring, or absent;
• wherein A, when present, is unsubstituted or substituted with one or more of halogen, OH, OCH3, OCH2CH3, OCH(CH3)2, OCH2CH(CH3)2, CH2OH, CH2OCH3, OCF3, OCH2CF3, NH2, CH2NH3, NHCH3, CH3, C2CH3, CHCH2, CH2CH(CH3)2, SO2CHCH2, NO2, amide, trihalomethyl, trihaloethyl, dihalomethyl, dihaloethyl, halomethyl, and haloethyl;
• wherein L is present or absent and is a 1-9 member alkyl, alkenyl, alkynyl, heteroalkyl, aminoalkyl, or amidealkyl chain optionally comprising one or more heteroatoms;
• wherein Y is alkylacrylamide, alkylacrylimide, propenal, 3-buten-2-one, acrylamide, vinyl sulfonamide, propynamide, ethyl propenoate, propenanamide, propenenitrile, nitroethylene, or absent;
• wherein R7 is H, D, halogen, OH, NH2, CH3.
