Abstract
Provided herein are stationary phase compositions comprising a chromatographic surface of porous or non-porous core material comprising a surface modifier for use in chromatographic separations.
Claims15
A reversed-phase chromatography method for the separation of one or more proteins from a sample, comprising:
- (i) applying the sample comprising one or more proteins to a stationary phase composition comprising a chromatographic surface of porous or non-porous core material comprising a surface modifier having the Formula I, II, III, or IV:
- or a combination thereof, wherein:
- x and y are each independently integers from 0 to 10;
- a is 0 to 5;
- b is 0 to 5;
- R1 and R2 are each independently selected from (C1-C3)alkyl, hydroxyl, fluorine, chlorine, bromine, cyano, —C(O)NH2, and carboxyl;
- R3 and R4 are each independently aryl optionally substituted with 1 to 5 groups selected from (C1-C3)alkyl, hydroxyl, fluorine, chlorine, bromine, cyano, —C(O)NH2, and carboxyl, or R3 and R4 are each independently (C1-C4)alkyl optionally substituted with cyano;
- R5 and R6 are each independently selected from hydrogen, trimethylsilyl, triethylsilyl, triisopropylsilyl, tert-butyldimethylsilyl, n-butyldimethylsilyl
- R7 is hydrogen, trimethylsilyl, triethylsilyl, triisopropylsilyl, tert-butyldimethylsilyl, or n-butyldimethylsilyl;
- and
- the hashed bond designates the point at which the surface modifier is attached, wherein the total phenyl surface coverage of the composition is greater than or equal to 5.3 μmol/m2;
- and
- (ii) eluting the sample through the stationary phase composition using a mobile phase, thereby separating the one or more proteins.

A reversed-phase chromatography method for the separation of oligonucleotides from a sample, comprising:
- applying the sample comprising oligonucleotides to a stationary phase composition comprising a chromatographic surface of porous or non-porous core material comprising a surface modifier having the Formula I, II, III, or IV:
- or a combination thereof, wherein:
- x and y are each independently integers from 0 to 10;
- a is 0 to 5;
- b is 0 to 5;
- R1 and R2 are each independently selected from (C1-C3) alkyl, hydroxyl, fluorine, chlorine, bromine, cyano, —C(O)NH2, and carboxyl;
- R3 and R4 are each independently aryl optionally substituted with 1 to 5 groups selected from (C1-C3) alkyl, hydroxyl, fluorine, chlorine, bromine, cyano, —C(O)NH2, and carboxyl, or R3 and R4 are each independently (C1-C4) alkyl optionally substituted with cyano;
- R5 and R6 are each independently selected from hydrogen, trimethylsilyl, triethylsilyl, triisopropylsilyl, tert-butyldimethylsilyl, n-butyldimethylsilyl,
- R7 is hydrogen, trimethylsilyl, triethylsilyl, triisopropylsilyl, tert-butyldimethylsilyl, or n-butyldimethylsilyl;
- and
- the hashed bond designates the point at which the surface modifier is attached, wherein the total phenyl surface coverage of the composition is greater than or equal to 5.3 μmol/m2;
- and
- (ii) eluting the sample through the stationary phase composition using a mobile phase, thereby separating the oligonucleotides.
