Abstract
A system for sorting a mixture of sperm cells in a fluid flow path, including stained sperm cells from two distinguishable groups, namely X chromosome bearing sperm and Y chromosome bearing sperm. The system and method can include an electromagnetic radiation source for exciting fluorescence emissions from the stained particles, a photodetector for detecting the fluorescence emissions from the stained particles, a processor for classifying the stained particles; and a photo-damaging laser for damaging selected particles in the flow path.
Claims17
A method of sorting a mixture of sperm cells contained in a fluid stream having either characteristic A or characteristic B comprising:
- a. delivering a fluid stream containing a stained population of sperm cells to a first location;
- b. interrogating the sperm cells at the first location with an interrogation laser that impinges each sperm cell that passes through the first location with multiple laser pulses and producing fluorescence emissions therefrom;
- c. detecting the fluorescence emissions produced by the sperm cells at the first location;
- d. classifying the sperm cells as having characteristic A or characteristic B from the detected fluorescence emissions;
- and
- e. selectively photo-damaging sperm cells in the fluid stream based on their classification.
The method of 5, wherein an amplitude of the force applied to the fluid stream is varied as a function of the break-off location.
