## Abstract

This disclosure is in the field of molecular biology and medical diagnosis and provides means and methods for determining the prognosis and disease outcome of a subject having a melanoma. More in particular, this disclosure provides a method for determining whether a subject having a melanoma has a poor prognosis, the method comprising the step of determining in a sample from the subject whether the LY75 promoter is methylated and if the LY75 promoter is methylated, classifying the subject as having a poor prognosis.

## Claims

### Claim 1

A method for treating a subject having melanoma with adjuvant therapy, the method comprising:
determining in a sample from the subject that the LY75 promoter is methylated; and then treating the subject with adjuvant therapy so as to treat the melanoma.

### Claim 2 (depends on 1)

The method according to claim 1, wherein the sample is obtained from the skin of the subject or from a melanoma.

### Claim 3 (depends on 1)

The method according to claim 1, wherein the sample is obtained from a biopsy taken from the subject.

### Claim 4 (depends on 1)

The method according to claim 1, further comprising determining tumor ulceration, determining metastatic disease at diagnosis, determining disease stage, and or determining Breslow thickness of the melanoma.

### Claim 5 (depends on 1)

The method according claim 1, wherein the methylation status of the LY75 promoter is determined by polymerase chain reaction-based methylation analysis.

### Claim 6 (depends on 5)

The method according to claim 5 wherein the polymerase chain reaction-based methylation analysis comprises nested primers.

### Claim 7 (depends on 1)

The method according to claim 1, wherein 20% or more of the LY75 promoters in the sample are methylated.
