A Myceliophthora thermophila filamentous fungal cell comprising a mutation that eliminates the activity of at least one endogenous metalloprotease, wherein the cell comprises a recombinant polynucleotide encoding a mammalian polypeptide, and wherein the endogenous metalloprotease has at least 50% identity to SEQ ID NO:
