Abstract
The present invention provides assays and assay systems for use in spatially encoded biological assays. The invention provides an assay system comprising an assay capable of high levels of multiplexing where reagents are provided to a biological sample in defined spatial patterns; instrumentation capable of controlled delivery of reagents according to the spatial patterns; and a decoding scheme providing a readout that is digital in nature.
Claims30
A method of detecting a target protein in a tissue sample, comprising:
- contacting a tissue sample with a plurality of probes, wherein a probe of the plurality of probes comprises an antibody that specifically binds a target protein of the tissue sample and is coupled to an oligonucleotide comprising a sequence;
- generating a nucleic acid molecule comprising all or a portion of the sequence of the oligonucleotide or a complement thereof and one or more nucleic acid tags that can be used to determine a location of the target protein in the tissue sample;
- and
- determining all or a portion of a sequence of the nucleic acid molecule or a complement thereof to detect the target protein and determine its associated location in the tissue sample.
