Abstract
The present invention relates to compositions comprising compounds for use in treating, ameliorating and/or preventing neuromuscular disorders. The compounds as defined herein preferably inhibit the ClC-1 ion channel. The invention further relates to methods of treating, preventing and/or ameliorating neuromuscular disorders, by administering said composition to a person in need thereof.
Claims28
A method of treating and/or ameliorating a neuromuscular disorder in a patient, comprising administering to a patient in need thereof a therapeutically effective amount of a compound of Formula (II):
- wherein
- A is selected from the group consisting of
- m is 0, 1 or 2;
- Y is selected from the group consisting of O, NH, N—CH3, CH2, CH2—O, CH2CH2—O, S, and SO2;
- X1, X2 and X3 are independently selected from CH or C(R4);
- R1 is H and R2 is selected from the group consisting of H, C1-4-alk(en)yl, C3-6-cycloalk(en)yl, wherein said C1-4-alk(en/yn)yl and C3-6-cycloalk(en)yl may be substituted with up to two substituents selected from the group consisting of —NR9—CO—R10, —N(R10)—SO2—R12, —CO—NR9R10, —SO2—NR9R10, —R13—O—R11, NR9R10, —S(O)R12, S(O)2R12, cyano, O—R11, fluorinated C1-3-alkyl, nitro and halo;
- or R1 and R2, together, form a C3-6-cycloalkyl R4 is selected from the group consisting H, halo, cyano, —CHO, C1-4alk(en)yl, halo-C1-4alk(en)yl, and —O—C1-4-alk(en)yl;
- R9, R10, R11 are independently selected from H or C1-4-alk(en/yn)yl and C3-6-cycloalk(en)yl;
- R12 is selected from C1-4-alk(en/yn)yl and C3-6-cycloalk(en)yl;
- R13 is selected from C1-4-alk(an/en/yn)diyl and C3-6-cycloalk(an/en)diyl;
- and
- R14 is C(O)OH;
- or a pharmaceutically acceptable salt, solvate, polymorph, or tautomer thereof, with the proviso that when A is
- m is 0, Y is O, X1 and X2 are CH, X3 is C(R4), and R2 is H, then R4 is not —O—C1-2-alkyl.
A method of reversing and/or ameliorating a neuromuscular blockade in a patient, comprising administering to a patient in need thereof a therapeutically effective amount of a compound of Formula (II):
- wherein
- A is selected from the group consisting of
- Y is selected from the group consisting of O, NH, N—CH3, CH2, CH2—O, CH2CH2—O, S, and SO2;
- X1, X2 and X3 are independently selected from CH or C(R4);
- R1 is H and R2 is selected from the group consisting of H, C1-4-alk(en)yl, C3-6-cycloalk(en)yl, wherein said C1-4-alk(en/yn)yl and C3-6-cycloalk(en)yl may be substituted with up to two substituents selected from the group consisting of —NR9—CO—R10, —N(R10)—SO2—R12, —CO—NR9R10, —SO2—NR9R10, —R13—O—R11, NR9R10, —S(O)R12, S(O)2R12, cyano, O—R11, fluorinated C1-3-alkyl, nitro and halo;
- or R1 and R2, together, form a C3-6-cycloalkyl R4 is selected from the group consisting H, halo, cyano, —CHO, C1-4alk(en)yl, halo-C1-4alk(en)yl, and —O—C1-4-alk(en)yl;
- R9, R10, R11 are independently selected from H or C1-4-alk(en/yn)yl and C3-6-cycloalk(en)yl;
- R12 is selected from C1-4-alk(en/yn)yl and C3-6-cycloalk(en)yl;
- R13 is selected from C1-4-alk(an/en/yn)diyl and C3-6-cycloalk(an/en)diyl;
- and
- R14 is C(O)OH;
- or a pharmaceutically acceptable salt, solvate, polymorph, or tautomer thereof, with the proviso that when A is
- m is 0, Y is 0, X1 and X2 are CH, X3 is C(R4), and R2 is H, then R4 is not —O—C1-2-alkyl.
