Abstract
The invention relates generally to compositions and methods for purifying the desired species from a mixture of desired heterodimer and contaminating homodimer immunoglobulin variants by modifying the isoelectric point(s) of the individual chains.
Claims3
A composition comprising a heterodimeric polypeptide comprising:
- a) a first variant human IgG1 heavy chain constant region comprising amino acid modifications as compared to a human IgG1 heavy chain constant region according to SEQ ID NO:
- 2, wherein said modifications comprise Q196K, 1199T, P217R, P228R, and N276K, according to the EU index as in Kabat;
- and
- b) a second variant human IgG1 heavy chain constant region comprising amino acid modifications as compared to a human IgG1 heavy chain constant region according to SEQ ID NO:
- 2, wherein said modifications comprise N203D, K274Q, R355Q, Q419E, and K447del, according to the EU index as in Kabat,
- and wherein said first variant human IgG1 heavy chain constant region and said second variant human IgG1 heavy chain constant region each possess at least 95% sequence identity to SEQ ID NO:
- 2.
