## Abstract

The invention relates to a recombinant baculoviral genome useful for the production of viral vectors for gene therapy, allowing said production from a single infection.

## Claims

### Claim 1

A recombinant baculovirus genome comprising:
(i) one or more expression cassettes comprising adeno-associated virus (AAV) rep and cap genes under the control of a promoter suitable for expression of said genes in an insect host cell, and
(ii) a recombinant genome of an AAV vector,
wherein said one or more expression cassettes and said recombinant genome of an AAV vector are each inserted into a locus selected from the egt locus and the polyhedrin locus.

### Claim 2 (depends on 1)

The recombinant baculovirus genome as claimed in claim 1, said rep and cap genes being contained in a single expression cassette.

### Claim 3 (depends on 1)

The recombinant baculovirus genome as claimed in claim 1, wherein said one or more expression cassettes and said recombinant genome of an AAV vector are each inserted into a different locus.

### Claim 4 (depends on 1)

The recombinant baculovirus genome as claimed in claim 1, wherein said one or more expression cassettes is(are) inserted into the egt locus.

### Claim 5 (depends on 1)

The recombinant baculovirus genome as claimed in claim 1, wherein said one or more expression cassettes is(are) inserted into the polyhedrin locus.

### Claim 6 (depends on 1)

The recombinant baculovirus genome as claimed in claim 1, wherein said recombinant genome of an AAV vector is inserted into the egt locus.

### Claim 7 (depends on 1)

The recombinant baculovirus genome as claimed in claim 1, wherein said recombinant genome of an AAV vector is inserted into the polyhedrin locus.

### Claim 8 (depends on 1)

The recombinant baculovirus genome as claimed in claim 1, wherein:
said one or more expression cassettes is(are) inserted into the polyhedrin locus; and
said recombinant genome of an AAV vector is inserted into the egt locus.

### Claim 9 (depends on 1)

The recombinant baculovirus genome as claimed in claim 1, wherein:
said one or more expression cassettes is(are) inserted into the egt locus; and
said recombinant genome of an AAV vector is inserted into the polyhedrin locus.

### Claim 10 (depends on 1)
