Abstract
An insulin conjugate having improved in vivo duration and stability is provided. The conjugate is prepared by covalently linking insulin with an immunoglobulin Fc region via a non-peptidyl polymer, a long-acting formulation comprising the same, and a preparation method thereof. The insulin conjugate of the present invention maintains in vivo activity of the peptide at a relatively high level and remarkably increases the serum half-life thereof, thereby greatly improving drug compliance upon insulin treatment.
Claims18
An insulin conjugate consisting of
- (i) an insulin moiety;
- (ii) a polyethylene glycol moiety of from about 3.4 kDa to less than 10 kDa in size;
- and
- (iii) an immunoglobulin Fc region moiety,
- wherein the insulin moiety is covalently linked at the N-terminal amino group of its B chain at one end of the polyethylene glycol moiety, and the other end of the polyethylene glycol moiety is covalently linked to the immunoglobulin Fc region moiety.
