Abstract
Provided herein is technology for gastrointestinal neoplasia screening and particularly, but not exclusively, to methods, compositions, and related uses for detecting the presence of gastrointestinal neoplasm, and classifying the site location of such a gastrointestinal neoplasm (e.g., a colorectal region, a pancreaticobiliary region, a gastroesophageal region).
Claims5
A method comprising:
- extracting genomic DNA from a biological sample of a human individual suspected of having or having lower gastrointestinal neoplasm or upper gastrointestinal neoplasm,
- treating the extracted genomic DNA with bisulfite,
- amplifying the bisulfite-treated genomic DNA using primers specific for a CpG site for QKI and primers specific for a CpG site for PDGFD, and
- measuring the methylation level of the CpG site for QKI and PDGFD by methylation-specific PCR, quantitative methylation-specific PCR, methylation-sensitive DNA restriction enzyme analysis, or bisulfite genomic sequencing PCR.
