Abstract
The present invention relates to a method for the mass-production of exosome comprising a cargo protein, a vector for preparing the exosome, exosome including a cargo protein prepared by the method, and a method for loading the cargo protein to cytosol by using the exosome prepared thereby. According to the method for preparing an exosome comprising a cargo protein provided by the present invention, the exosome loaded with a cargo protein can be produced with a high yield, so that it can be used broadly in the treatment of disease using the exosome.
Claims17
A method for mass production of an exosome containing a cargo protein comprising:
- introducing a polynucleotide encoding a fusion protein composed of an exosome specific marker and a first photo-specific binding protein (fusion protein I) and a polynucleotide encoding a fusion protein composed of a cargo protein and a second photo-specific binding protein that can be linked to the first photo-specific binding protein (fusion protein II) into an exosome production cell, thereby transforming the cell;
- and
- b) culturing the transformed cell of step a).
A vector composition for production of an exosome comprising:
- (a) a first expression vector containing a polynucleotide encoding a fusion protein composed of an exosome specific marker and a first photo-specific binding protein (fusion protein I);
- and
- (b) a second expression vector containing a multicloning site and a polynucleotide encoding a second photo-specific binding protein to be linked to the first photo-specific binding protein.
Transformed cells produced by a method comprising:
- a) introducing a polynucleotide encoding a fusion protein composed of an exosome specific marker and a first photo-specific binding protein (fusion protein I) and a polynucleotide encoding a fusion protein composed of a cargo protein and a second photo-specific binding protein that can be linked to the first photo-specific binding protein (fusion protein II) into an exosome production cell.
