Abstract
A compound can be a pro-fluorophore peroxynitrite sensor that generates a fluorophore when cleaved by peroxynitrite, having a structure of Formula A: wherein: moiety A is an ER-targeting fluorophore; Y is a linker; and moiety B is a phenol, substituted or unsubstituted, wherein the structure of Formula A is less fluorescent than the ER-targeting fluorophore moiety A.

A method of detecting peroxynitrite comprising:
- providing a compound comprising a structure of Formula A:
- wherein:
- moiety A is an endoplasmic reticulum-targeting fluorophore;
- Y is a linker;
- and
- moiety B is a phenol, substituted or unsubstituted,
- wherein the structure of Formula A is less fluorescent than the endoplasmic reticulum-targeting fluorophore moiety A;
- contacting the compound with the endoplasmic reticulum;
- allowing peroxynitrite to cleave the compound into a fluorophore and a p-benzoquinone when present;
- and
- detecting the fluorescence of the fluorophore.
