Abstract
Provided herein are multiplex assays for detecting antibodies indicative of presence and stage of syphilis infection in an individual. Individuals infected with syphilis produce antibodies directed to syphilis components and the lipid cellular debris associated with the infection. The present disclosure represents the first combination of these diverse antibody targets in a single assay.
Claims14
A method of detecting the presence of syphilis antibodies in a sample comprising:
- (a) contacting the sample with a mixture comprising (i) beads non-covalently attached to cardiolipin-lecithin-cholesterol antigen, wherein the beads are prepared by incubating cationic beads with the cardiolipin-lecithin-cholesterol antigen in a coupling buffer to coat the beads with cardiolipin-lecithin-cholesterol by ionic interaction;
- and (ii) beads coated with Treponema pallidum antigen, thereby forming a sample-bead mixture;
- (b) washing the sample-bead mixture to remove unbound sample;
- (c) contacting the sample-bead mixture with a labeled, anti-human immunoglobulin (Ig) antibody;
- and
- (d) detecting the presence of syphilis antibodies by detecting the label on the anti-human Ig antibody.
