Abstract
The present disclosure provides compositions and methods for efficient and effective protein delivery in vitro and in vivo. In some aspects, proteins are reversibly crosslinked to each other and/or modified with functional groups and protected from protease degradation by a polymer-based or silica-based nanoshell.

A method for treating cancer by stimulating or enhancing an immune response in a human subject, the method comprising:
- administering to the human subject a composition comprising
- (i) an expanded population of lymphocytes specific to more than one tumor antigen;
- and
- (ii) a nanostructure conjugated to the surface of each lymphocyte, the nanostructure comprising a plurality of immunostimulatory proteins reversibly crosslinked to each other through a degradable linker that degrades under physiological conditions to release the immunostimulatory proteins,
- wherein the immunostimulatory proteins stimulate or enhance an immune response in the human subject, thereby treating cancer.
A method for treating cancer by stimulating or enhancing an immune response in a human subject, the method comprising:
- administering to the human subject a composition comprising
- (i) a population of lymphocytes specific to at least one tumor antigen;
- and
- (ii) a nanostructure conjugated to the surface of each lymphocyte, the nanostructure comprising a plurality of immunostimulatory proteins reversibly crosslinked to each other through a degradable linker that degrades under physiological conditions to release the immunostimulatory proteins,
- wherein the immunostimulatory proteins stimulate or enhance an immune response in the human subject, thereby treating cancer.
An immunostimulatory composition for treating cancer by stimulating or enhancing an immune response in a human subject, the composition comprising:
