Abstract
The present disclosure relates to engineered cells that include genetic alterations leading to up- or down-regulation of certain genes in the cells for improved production of a recombinant protein. Also provided are methods of preparing and using such cells.
Claims11
An isolated engineered mammalian cell comprising one or more genetic alterations resulting in increased expression or activity of hk1 (hexokinase 1), MAPK3 (mitogen-activated protein kinase 3), CAP (catabolite activator protein) and Nrf2 (nuclear factor (erythroid-derived 2)-like 2), wherein the increase is as compared to a corresponding mammalian cell without the genetic alterations.
An isolated mammalian cell comprising one or more exogenous polynucleotides encoding hk1 (hexokinase 1), MAPK3 (mitogen-activated protein kinase 3), CAP (catabolite activator protein) and Nrf2 (nuclear factor (erythroid-derived 2)-like 2).
