Abstract
The present invention relates to alpha-amylase variants. The present invention also relates to polynucleotides encoding the variants; nucleic acid constructs, vectors, and host cells comprising the polynucleotides; and methods of using the variants.
Claims23
An alpha-amylase variant comprising
- a) a deletion at two or more positions corresponding to positions R180, S181, T182 and G183 of the mature polypeptide of SEQ ID NO:
- 1 or 2, and
- b) an alteration at two or more positions corresponding to positions Y48, E169, S170, R171, K172, N174, M317, I390, of the mature polypeptide of SEQ ID NO:
- 1 or 2, wherein each alteration is independently a substitution or an insertion,
- wherein the variant has at least 80%, but less than 100%, sequence identity, with the mature polypeptide of SEQ ID NO:
- 1 or 2, wherein the variant has alpha-amylase activity.

A method for obtaining an alpha-amylase variant, comprising introducing into a parent alpha-amylase having at least 80% sequence identity to SEQ ID NO:
- 1 or 2
- a) a deletion at two or more positions corresponding to positions R180, S181, T182 and G183 of the mature polypeptide of SEQ ID NO:
- 1 or 2 and
- b) an alteration at two or more positions corresponding to positions Y48, E169, S170, R171, K172, L173, N174, L205, R309, M317, I390 of the mature polypeptide of SEQ ID NO:
- 1 or 2, wherein each alteration is independently a substitution or insertion,
- wherein the variant has at least 80%, but less than 100%, sequence identity with the mature polypeptide of SEQ ID NO:
- 1 or 2, wherein the variant has alpha-amylase activity;
- and recovering the variant.

A method of improving the detergent stability of an alpha-amylase by introducing into a parent alpha-amylase
- a) a deletion at two or more positions corresponding to positions R180, S181, T182 and G183 of the mature polypeptide of SEQ ID NO:
- 1 or 2 and
- b) an alteration at two or more positions corresponding to positions Y48, E169, S170, R171, K172, N174, M317, I390 of the mature polypeptide of SEQ ID NO:
- 1 or 2, wherein each alteration is independently a substitution or insertion,
- wherein the resulting variant has at least 80%, but less than 100%, sequence identity with the mature polypeptide of SEQ ID NO:
- 1 or 2, wherein the resulting variant has alpha-amylase activity and an improved detergent stability compared to the parent alpha-amylase.
