Abstract
The present invention relates to antibodies specific for pyroglutamated Aβ, as well as their use in the treatment of Alzheimer's disease and as use in diagnostic methods or as diagnostic imaging ligands. Further, is provided pyroglutamated N-terminal fragments of murine or human Aβ to generate antibodies and for use in therapeutic purposes.
Claims12
An IgG antibody, or a fragment thereof, wherein said antibody and said fragment are capable of specifically binding murine AβpE3 with a binding affinity (KD) of about 100 nM to about 5 nm for the murine pyroglutamated Aβ fragment pEFGHDSGFE (SEQ ID NO:68), wherein said antibody comprises:
- (A) a heavy chain variable region (VH) CDR1 comprising GLTLSDAWMN (SEQ ID NO:32) or GITLNDAWMT (SEQ ID NO:38);
- (B) a heavy chain variable region (VH) CDR2 comprising EIRSKAYKHATYYAESVKG (SEQ ID NO:33) or EIRNKANNHATNYAESVKG (SEQ ID NO:39);
- (C) a heavy chain variable region (VH) CDR3 comprising NO:64) HGS (SEQ ID NO:34) or HSY (SEQ ID NO:40);
- (D) a light chain variable region (VL) CDR1 comprising RASQGISSKMG (SEQ ID NO:35) or HASQGIRNNIG (SEQ ID NO:41);
- (E) a light chain variable region (VL) CDR2 comprising HGTKLED (SEQ ID NO:36) or HGTKLED (SEQ ID NO:42);
- and
- (F) a light chain variable region (VL) CDR3 comprising VQYAQFPYT (SEQ ID NO:37) or VQYDQFPYT (SEQ ID NO:43).
