Abstract
The present disclosure relates to compositions and methods useful for the production of heterologous proteins in filamentous fungal cells.
Claims9
A Trichoderma reesei filamentous fungal cell comprising at least one endogenous protease having reduced or no protease activity, and a recombinant polynucleotide encoding a heterologous polypeptide, wherein the gene encoding endogenous amp2 protease of SEQ ID NO:775 comprises a mutation that reduces or eliminates the activity of the endogenous amp2 protease, and wherein the production level of the heterologous polypeptide is at least two-fold higher than the production level of the same polypeptide as produced in a corresponding parental filamentous fungal cell in which the gene encoding endogenous amp2 protease of SEQ ID NO:775 does not comprise a mutation that reduces or eliminates the activity of the endogenous amp2 protease.
