Abstract
The present invention relates to mixtures comprising a polypeptide or a plurality of polypeptides having biomass-degrading activity that is solubilized from an inclusion body, and retaining biomass-degrading activity, and methods for producing and using the same. The invention described herein provides methods for increasing the yield of recombinant protein with biomass-degrading activity that can be isolated from host cells.
Claims31
A mixture comprising a plurality of polypeptides having biomass-degrading activity and a chaotropic agent, wherein:
- (i) the polypeptides have at least 20-40% of the biomass-degrading activity compared to a native polypeptide having biomass-degrading activity in the absence of the chaotropic agent;
- (ii) the plurality comprises a polypeptide comprising the amino acid sequence of SEQ ID NO:
- 1 and having cellobiase activity;
- and
- (iii) the chaotropic agent is selected from urea, a substituted urea, or guanidinium hydrochloride.
A mixture comprising a plurality of polypeptides having biomass-degrading activity and a solubilizing agent, wherein the polypeptides have at least 8-10% of the biomass-degrading activity compared to a native polypeptide having biomass-degrading activity and the plurality comprises a polypeptide having the amino acid sequence of SEQ ID NO:
- 1 having cellobiase activity, wherein the plurality of polypeptides having biomass-degrading activity is solubilized from inclusion bodies.
