Abstract
The present invention provides modified oocytes having a nuclear genome derived from a first oocyte and cytoplasm derived from a second oocyte from a different subject, and methods for making and using such modified oocytes. The methods and compositions of the present invention can be useful in a variety of settings including, but not limited to, in in vitro fertilization (“IVF”) procedures.
Claims29
A method of reducing transmission of a mitochondrial DNA mutation from a mother having the mitochondrial DNA mutation to the mother's offspring, the method comprising using a modified oocyte in an assisted reproduction procedure, wherein the modified oocyte is produced by a method comprising:
- introducing a nuclear genome from a first oocyte having the mitochondrial DNA mutation into an enucleated second oocyte not having the mitochondrial DNA mutation to form the modified oocyte, wherein the nuclear genome is from a metaphase I oocyte or an immature metaphase II oocyte, and wherein the first oocyte and the second enucleated oocyte are mammalian and from mammals of the same species, thereby preventing or reducing transmission of the mitochondrial DNA mutation.
A method of reducing transmission of a mitochondrial DNA mutation from a mother having the mitochondrial DNA mutation to the mother's offspring, the method comprising using a modified oocyte in an assisted reproduction procedure, wherein the modified oocyte is produced by a method comprising:
- a) treating a karyoplast comprising a nuclear genome from a first oocyte having the mitochondrial DNA mutation to induce partial spindle depolymerization, wherein the nuclear genome is a mature metaphase II nuclear genome;
- and
- b) introducing the karyoplast comprising the partially depolymerized spindle into an enucleated second oocyte not having the mitochondrial DNA mutation, thereby producing the modified oocyte, wherein the first oocyte and the second enucleated oocyte are mammalian and from animals mammals of the same species, thereby reducing transmission of the mitochondrial DNA mutation.
