Abstract
The inventive compounds are small molecule therapeutics that are potent inhibitors of arginase 1 and arginase 2 activity. The inventions also provides pharmaceutical compositions and methods for using the inventive compounds for treating or preventing a disease or condition associated with arginase activity.

A compound represented by formula (I), or a stereoisomer, a tautomer, a pharmaceutically acceptable salt, or a solvate thereof:
- wherein:
- U is —C(═NH)—, —S(O)—, or —S(O)2—;
- W is —CO2H, —C(O)O((C1-C6)alkyl), —C(O)NHOH, —C(O)NHCN, —C(O)NH2, —C(O)NH((C1-C6)alkyl), —C(O)N((C1-C6)alkyl)2, —C(O)NH(aryl), —C(O)N(aryl)((C1-C6)alkyl), —C(O)N(aryl)2, —C(O)NH((C1-C6)haloalkyl), —S(O)2NH2, —S(O)2NH((C1-C6)alkyl), —S(O)2NH((C1-C6)haloalkyl), —S(O)2NH(aryl), —S(O)2NHC(O)((C1-C6)alkyl), —S(O)2NHC(O)((C1-C6)haloalkyl), —S(O)2NHC(O)(aryl), —N(H)S(O)2((C1-C6)alkyl), —N(H)S(O)2(aryl), N(H)S(O)2((C1-C6)haloalkyl), —NHC(O)((C1-C6)alkyl), —NHC(O)((C1-C6)haloalkyl), —NHC(O)(aryl), —NHC(O)NH((C1-C6)alkyl), —NHC(O)NH(aryl), —C(O)N(H)S(O)2((C1-C6)alkyl), —C(O)N(H)S(O)2(aryl), —C(O)N(H)S(O)2((C1-C6)haloalkyl), —P(O)(OH)2, —CF3,
- X is —N(R)1— or —C(R)2—;
- Y is —(CR)2—;
- Z is —C(R)2—, —O—, —S—, —SO—, or —SO2—;
- each R independently is selected from —H, —(C1-C3)alkyl, -halo, or —OH;
- R1 is —H or —SO2NH2 or —(C1-C3)alkyl;
- each R2 independently is selected from H, alkyl, aryl, arylalkyl, heteroaryl, heteroarylalkyl, heterocyclyl, or heterocyclylalkyl;
- R3 and R4 are each independently selected from hydrogen, straight or branched (C1-C6)alkyl, and C(O)—R′,
- or R3 and R4 together with the boron atom to which they are bound via oxygen atoms form a 4-, 5-, 6-, or 7-membered ring that is fully saturated, or partially saturated.
