Abstract
The invention relates to a cell capable of converting one or more pentose sugar and one or more hexose sugar into fermentation product constitutively expressing one or more heterologous or homologous polypeptide having the amino acid sequence set out in SEQ ID NO: 20, or a variant polypeptide thereof having at least 45% identity to SEQ ID NO 20. In an embodiment the heterologous polypeptide has glyoxalase activity.
Claims19
A Saccharomyces cell capable of converting one or more pentose sugar and one or more hexose sugar into fermentation product, comprising a nucleotide sequence encoding a xylose isomerase, said cell constitutively expressing one or more heterologous or homologous polypeptide-having the amino acid sequence set out in SEQ ID NO:20, or a variant polypeptide thereof having at least about 60% sequence identity to SEQ ID NO:20, wherein the coding region of the GRE3 aldose reductase gene is inactivated, and wherein the sugar conversion is faster as compared to a control Saccharomyces which does not constitutively express the polypeptide of SEQ ID NO:
- 20 or a variant thereof having at least about 60% sequence identity to SEQ ID NO:
- 20.
A Saccharomyces cell capable of converting one or more pentose sugar and or one or more hexose sugar into fermentation product, comprising a constitutively expressed heterologous or homologous polynucleotide which comprises:
- (a) the nucleotide sequence as set forth in SEQ ID NO:27;
- or
- (b) codon-pair optimized nucleotide sequence of SEQ ID NO:27, wherein the coding region of the GRE3 aldose reductase gene is inactivated, and wherein the sugar conversion is faster as compared to a control Saccharomyces which does not comprise SEQ ID NO:
- 27, or a codon-pair optimized sequence thereof, and which does not contain an inactivated GRE3 aldose reductase gene.
A Saccharomyces cell capable of converting one or more pentose sugar and one or more hexose sugar into fermentation product under micro-aerophilic or anaerobic conditions, comprising a nucleotide sequence encoding a xylose isomerase, said cell constitutively expressing one or more heterologous or homologous polypeptide-having the amino acid sequence set out in SEQ ID NO:20, or a variant polypeptide thereof having at least about 60% sequence identity to SEQ ID NO:20, wherein the coding region of the GRE3 aldose reductase gene is inactivated, and wherein the sugar conversion is faster as compared to a control Saccharomyces which does not constitutively express the polypeptide of SEQ ID NO:
- 20 or a variant thereof having at least about 60% sequence identity to SEQ ID NO:
- 20, cultured under the same micro-aerophilic or anaerobic conditions.
