Abstract
The invention relates to ketoreductases and the use thereof. The ketoreductases of the invention are particularly useful for enzymatically catalyzing the reduction of ketones to chiral secondary alcohols.
Claims20
A ketoreductase comprising an amino acid sequence with at least 72% homology to SEQ ID NO:2, wherein SEQ ID NO:
- 2 is engineered in at least one or more positions selected from the group consisting of the positions Y21, V23, S33, L39, R40, A43, P68, V89, G95, P97, T98, D103, G109, V119, L121, V124, Y125, I149, L150, S154, E155, T157, A158, T163, H190, Y193, L198, L199, A201, A206, Y207, V229, and V247;
- wherein the ketoreductase exhibits improved specific activity, temperature stability and/or stereoselectivity compared to the polypeptide of SEQ ID NO:
- 2.
