Abstract
Bifunctional compounds that increase uric acid excretion and reduce uric acid production, and monofunctional compounds that either increase uric acid excretion or reduce uric acid production. Methods of using these compounds for reducing uric acid levels in blood or serum, for treating disorders or uric acid metabolism, and for maintaining normal uric acid levels in blood or serum are provided. Pharmaceutical compositions comprising the bifunctional and monofunctional compounds are also provided.

A compound selected from the group consisting of
- a) compounds having a structure represented by Formula (I):
- wherein
- X is O or S;
- W is present or absent, and if present is one or more hydroxyl moieties, R1OH, or at least one hydroxyl moiety and R1OH;
- and
- Z is present or absent, and if present is SO2N(R2)2, —R1CO2H,
- —NR2SO2R3, —NR2SO2N(R2)2, or —NR2C(O)N(R2)2;
- wherein R1 is alkyl;
- wherein each R2 is independently H, alkyl or aryl, each optionally substituted with one or more halogen atoms or OR2;
- wherein each R3 is independently alkyl or aryl, each optionally substituted with one or more halogen atoms or OR2;
- and
- wherein each Xa is independently selected from either O or S,
- provided that at least one of W and Z is present, and if W is present and Z is absent, W is not 4′-hydroxy;
- b) compounds having a structure represented by Formula (II):
- wherein
- X is O or S;
- and
- each Z is independently present or absent and, if present, is independently selected from one or more halogen atoms, —CN, —CF3, —OR2, —C(O)R2, SR2, —S(O)fR3 where f is 1 or 2, —N(R2)2, —NO2, —CO2R2, —OCO2R3, OC(O)R2, —CON(R2)2, —NR2C(O)R2, —SO2(NR2)2, —NR2SO2R3, —NR2SO2N(R2)2, —NR2C(O)N(R2)2, alkyl, aryl, alkenyl and alkynyl;
- wherein each R2 is independently H, alkyl or aryl;
- wherein each R3 is independently alkyl or aryl, optionally substituted with one or more halogen atoms or OR2;
- and
- wherein a, b, c, d, and e are each independently carbon or nitrogen, with the proviso that at least one of a, b, c, d and e is nitrogen and Z is not connected directly to a nitrogen, except that Z may optionally be connected to a nitrogen at a, b, c, d, or e by replacement of the hydrogen of an NH group when Z is —C(O)R2, —S(O)fR3, —CO2R3, —CON(R2)2, —SO2N(R2)2, alkyl, aryl, alkenyl or alkynyl;
- c) compounds having a structure represented by Formula (III):
- wherein
- X is O or S;
- and
- each Z is independently present or absent and, if present, is independently selected from one or more halogen atoms, —CN, —CF3, —OR2, —C(O)R2, SR2, —S(O)fR3 where f is 1 or 2, —N(R2)2, —NO2, —CO2R2, —OCO2R3, OC(O)R2, —CON(R2)2, —NR2C(O)R2, —SO2(NR2)2, —NR2SO2R3, —NR2SO2N(R2)2, —NR2C(O)N(R2)2, alkyl, aryl, alkenyl and alkynyl;
- wherein each R2 is independently H, alkyl or aryl;
- wherein each R3 is independently alkyl or aryl, optionally substituted with one or more halogen atoms or OR2;
- and
- wherein a, b, and c, are each independently carbon or nitrogen, with the proviso that at least one of a, b, and c is nitrogen and Z is not connected directly to a nitrogen, except that Z may optionally be connected to a nitrogen at a, b, or c by replacement of the hydrogen of an NH group when Z is —C(O)R2, —S(O)fR3, —CO2R2, —CON(R2)2, —SO2N(R2)2, alkyl, aryl, alkenyl or alkynyl;
