Abstract
The invention provides for modified reovirus nucleic acid sequences and modified reovirus polypeptide sequences as well as reoviruses containing such modified nucleic acid or polypeptide sequences. The invention also provides for pharmaceutical compositions that include reoviruses having a modified sequence as well as methods of making and using such reoviruses.
Claims27
A modified type 3 reovirus comprising a type 3 reovirus lambda-3 polypeptide, wherein the type 3 reovirus lambda-3 polypeptide comprises SEQ ID NO:23 with one or more amino acid modifications, wherein the one or more amino acid modifications are selected from the group consisting of an Ala at residue 267, a Thr at residue 557, a Lys at residue 755, a Met at residue 756, a Pro at residue 926, a Pro at residue 963, a Leu at residue 979, and an Arg at residue 1045, or any combination thereof, numbered relative to SEQ ID NO:23 (GenBank Accession No. M24734.1).
A reovirus comprising a lambda-3 polypeptide having an amino acid modification which is a Leu at residue 979, an Arg at residue 1045 or a combination thereof, and optionally one or more further amino acid modifications, wherein the one or more further amino acid modifications are selected from the group consisting of a Val at residue 214, an Ala at residue 267, a Thr at residue 557, a Lys at residue 755, a Met at residue 756, a Pro at residue 926, a Pro at residue 963, a Val at residue 1071, or any combination thereof, numbered relative to SEQ ID NO:23 (GenBank Accession No. M24734.1),
- wherein the reovirus exhibits a growth advantage over a reovirus that does not contain a corresponding modification or modifications.
